    The Critical Aggregation Concentration of ß-Lactoglobulin-Based Fibril Formation

    The critical aggregation concentration (CAC) for fibril formation of ß-lactoglobulin (ß-lg) at pH 2 was determined at 343, 353, 358, 363, and 383 K using a Thioflavin T assay and was approximately 0.16 wt%. The accuracy of the CAC was increased by measuring the conversion into fibrils at different stirring speeds. The corresponding binding energy per mol, as determined from the CAC, was 13 RT (~40 kJ mol¿1) for the measured temperature range. The fact that the CAC was independent of temperature within the experimental error indicates that the fibril formation of ß-lg at pH 2 and the measured temperature range is an entropy-driven process

    Gluon confinement criterion in QCD

    We fix exactly and uniquely the infrared structure of the full gluon propagator in QCD, not solving explicitly the corresponding dynamical equation of motion. By construction, this structure is an infinite sum over all possible severe (i.e., more singular than 1/q21/q^2) infrared singularities. It reflects the zero momentum modes enhancement effect in the true QCD vacuum, which is due to the self-interaction of massless gluons. It existence automatically exhibits a characteristic mass (the so-called mass gap). It is responsible for the scale of nonperturbative dynamics in the true QCD ground state. The theory of distributions, complemented by the dimensional regularization method, allows one to put the severe infrared singularities under the firm mathematical control. By an infrared renormalization of a mass gap only, the infrared structure of the full gluon propagator is exactly reduced to the simplest severe infrared singularity, the famous (q2)2(q^2)^{-2}. Thus we have exactly established the interaction between quarks (concerning its pure gluon (i.e., nonlinear) contribution) up to its unimportant perturbative part. This also makes it possible for the first time to formulate the gluon confinement criterion and intrinsically nonperturbative phase in QCD in a manifestly gauge-invariant ways.Comment: 10 pages, no figures, no tables.     The efforts of direct support professionals to facilitate inclusion:The role of psychological determinants and work setting

    BackgroundVarious studies have found that direct support professionals (DSPs) play an important role in determining the degree to which people with intellectual disabilities (ID) are included in society. However, less research has been conducted on the psychological processes that may influence the behavioural intentions of DSPs to actually engage with and invest effort in supporting their clients' inclusion. Five possible psychological variables are identified in the literature: attitudes, social norms, experienced competencies, identity and meta‐evaluation. In our research, we tested whether these processes influence the (intended) efforts DSPs make to facilitate their clients' inclusion.MethodA structured questionnaire was sent to 927 DSPs working in one of three different locations (an ordinary non‐segregated setting, a reversed non‐segregated setting and a residential facility). Of these, 336 DSPs completed the questionnaire.ResultsSeveral variables revealed differences between the three locations, specifically in efforts to facilitate inclusion, attitudes, social norms, experienced competencies and professional identity. Looking at the overall means, we found (relatively) high scores for the experienced competencies, role identity and meta‐evaluation. In contrast, the means were relatively negative regarding the DSPs' attitudes to inclusion and their assumed social norms.ConclusionsDirect support professionals' efforts to facilitate inclusion depend on their attitude towards inclusion, the experienced competencies, their role identity, the DSPs' meta‐evaluation and, indirectly through attitudes, also on the assumed social norms of the relevant stakeholders. Organizations responsible for supporting people with ID and which may want their DSPs to make greater efforts to facilitate inclusion should pay attention to these psychological variables

    Enzyme-induced Formation of ß-Lactoglobulin Fibrils by AspN Endoproteinase

    This paper describes a low temperature, enzymatic route to induce fibrillar structures in a protein solution. The route comprises two steps. First, ß-lactoglobulin was hydrolyzed into peptides at pH 8 and 37°C with the enzyme AspN endoproteinase, which resulted in the formation of random aggregates. After hydrolysis, the pH was lowered to 2. As a result, long fibrillar aggregates were formed which was observed using transmission electron microscopy and Thioflavin T fluorescence measurements

    Heterologous Processing and Export of the Bacteriocins Pediocin PA-1 and Lactococcin A in Lactococcus Lactis:A Study with Leader Exchange

    The bacteriocins pediocin PA-1 and lactococcin A are synthesized as precursors carrying N-terminal extensions with a conserved cleavage site preceded by two glycine residues in positions -2 and -1. Each bacteriocin is translocated through the cytoplasmic membrane by an integral membrane protein of the ABC cassette superfamily which, in the case of pediocin PA-1, has been shown to possess peptidase activity responsible for proteolytic cleavage of the pre-bacteriocin. In each case, another integral membrane protein is essential for bacteriocin production. In this study, a two-step PCR approach was used to permutate the leaders of pediocin PA-1 and lactococcin A. Wild-type and chimeric pre-bacteriocins were assayed for maturation by the processing/export machinery of pediocin PA-1 and lactococcin A. The results show that pediocin PA-1 can be efficiently exported by the lactococcin machinery whether it carries the lactococcin or the pediocin leader. It can also compete with wild-type lactococcin A for the lactococcin machinery. Pediocin PA-1 carrying the lactococcin A leader or lactococcin A carrying that of pediocin PA-1 was poorly secreted when complemented with the pediocin PA-1 machinery, showing that the pediocin machinery is more specific for its bacteriocin substrate. Wild-type pre-pediocin and chimeric pre-pediocin were shown to be processed by the lactococcin machinery at or near the double-glycine cleavage site. These results show the potential of the lactococcin LcnC/LcnD machinery as a maturation system for peptides carrying double-glycine-type amino-terminal leaders

    Post‐transplant inotrope score is associated with clinical outcomes after adult heart transplantation

    Background: Inotrope score has been proposed as a marker of clinical outcome after adult heart transplantation (HTx) but is rarely used in practice. Methods: Inotrope score during the first 48 h after HTx was calculated in 81 patients as: dopamine + dobutamine + amrinone + milrinone (dose × 15) + epinephrine (dose × 100) + norepinephrine (dose × 100) + enoximone + isoprenaline (dose × 100), with each drug in µg/kg/min. Determinants of inotrope score were identified with linear regression. Cox regression was used to determine the association of inotrope score with mortality. Results: The mean recipient age was 52 ± 11 years, and 32 (39.5%) patients were female. Determinants of inotrope score were preoperative C-reactive protein, serum urea, congenital heart disease, and donor cardiac arrest (R2 = .30). Inotrope score was associated with 5-year mortality, independent of recipient age and gender (HR 1.03, 95% CI 1.00-1.07). This association was attenuated when adjusting for female-to-male transplant and ischemia time. Inotrope score was also strongly associated with continuous veno-venous hemofiltration (OR 1.07, 95% CI 1.03-1.12). Conclusion: High inotrope score post-HTx was observed in recipient congenital heart disease and was associated with a higher risk of mortality and acute kidney injury

    The aim of this Letter to the Editor was to report some methodological shortcomings in a recently published article. Issues regarding missing values and overfitting are mentioned. First, Complete Case (CC) analysis was used instead of an imputation method. Second, there was a high chance of overfitting and lack of model validation. In conclusion, the results of this study should be interpret with caution and further research is necessary
